My boss will be retiring in about two years. Over the weekend, I decided I want her job. The question of the day is: when do I start laying groundwork and making murmurings about this?
A. Now, to mark your territory and discourage other applicants.
B. A year from now.
C. When the official vacancy is published.
D. Never, you don’t really want this job. You are, at heart, a lazy ass.

Sort of A. Discuss first with mentor, which bonus if that person IS your boss. See what areas you need to improve and any training you need to get. Do that then warn all those other worker bees off. Helps to be qualified and have that desire for the job YMMV.
I was going to answer B; however, llj above makes some valid points.
My only concern would be the following: has your boss made it known to everyone that she will be retiring?...or did she just mention it to you? If it's a given that she's going and no big secret, then there's no problem going up to her and doing what llj proposes...and yet, at the same time, two years away strikes me as somewhat cut-throat...Depends on the relation you have with your boss.

My boss has made it very clear she is going at 60. She HAS to retire. And maybe even earlier. She could take early retirement now, if she wanted to. So everyone knows.
There is a possibility her job could be abolished. They are doing that with a lot of people, but on the chance that it won't be, I am thinking how to position myself.
This is a cut-throat place. Maybe it is good to be the throat-cutter for a change instead of the throat-cuttee, which I have been in the past.
Oh, yes, I need the advice of the evil Invisible Man. He would know how to screw someone over and get the job.

However, first you need to work out why you want the job- is it the work? The money? The prestige? The office?
If you still want the job, then its definately A.
